What does it mean when an agent asks....

[FONT=Arial, Helvetica, sans-serif]I get why agents ask whether other agents are currently reading the manuscript requested. But what I don't get is the second question some ask: whether other agents have read the manuscript before.

Thoughts?

Because they want to know who expressed interest in your book, and who has already turned it down. It gives them a bit of insight, that's all.

You don't have to tell them, but you can if you want.
